Can I Use Back-to-Source by Mirroring to Access an Internal Service or Private IP Address?

No.

A back-to-source by mirroring rule allows only the origin server to be configured as a publicly accessible domain name or public IP address.

If you want to configure a service within a VPC as the origin server, use a NAT gateway or ELB to make its domain name accessible from the public network.
